how can i calculate x,y cordinates of an area of interest from an image in matlab.

It depends on how you create your ROI. Are you using imrect(), rbbox(), imfreehand(), thresholding, color segmentation, or some other method? It depends on which method you use and if you need just the coordinates of the boundary/outline of it (which is the usual case), or if you need the coordinates of every pixel inside the ROI (usually not needed and not the most efficient approach).
