Optimisation non-linear constraints

I have a optimization issue where I would like to minimize
V = (q*(c1i-x(3)))./(x(3)*kf(x(1))-x(2)*kr(x(1)));
I have 3 variables ( x(1), x(2), x(3) )
- The initial condition xo is it only for the three variables or it includes also the the function V
So, should the initial value vector of 3 or 4 elements
x0 = [X1 X2 X3]' OR x0 = [V X1 X2 X3]
2. x(2) have a linre non-equality constraint ( X(2) <= 2), How can let enter this contraints as I have three variables
