imfill for the inside of the object

I have an image with one giant holes(contour) which is a white pixels edge and inside it a white pupil. I need your help trying to fill the one giant hole(iris) so that I can analyze just that one. I've tried using the imfill function but so far it has been unsuccessful i used Ifill = imfill(I,'holes'); and tried a seed in the hole as imfill(I,[300 382]) .(i dont know how to upload my images to matlab central for you to see) any help on this subject would be greatly appreciated. Thanks yall.

Note that I must be type logical, or else the hole you are trying to fill must have intensity=zero exactly.

This is a later post of http://www.mathworks.com/matlabcentral/answers/51406-imfill-inside-an-hole, which you should have continued. But now I'm going to delete that one and transfer my answer from your fist question here. In the future, don't make multiple posts for the same question.

I use imfill all the time. Why was it unsuccessful? Did you create a binary image of your outer object? Then, didn't imfill fill in the hole? Please upload your original image and your binary image, and your imfill code.
