That plots one point but by default the line created by plot includes no Marker.
axis([0 1 0 1])
hold on
h = plot(0.25, 0.25); % no line shown, no marker
h.Marker % 'none'
You can specify a Marker when you create the plot.
h2 = plot(0.75, 0.75, 'o'); % One circle marker shown
Or if you have a handle to the line that was created by plot you can change its properties.
h.Marker = 's'; % now one circle marker and one square marker shown
