Create a matrix from x y coordinates

Hello,
I have an array with two columns of x and y coordinates (values are in cm). From this array I would like to create a square matrix of 256 x 256 elements (which represent a matrix of 256 x 256 pixels). Every pixel must have a dimension of 5 x 5 cm². As for elements, they should represent the number of points (xi,yi) inside of the pixel.
I tried a lot of ways, but none of these is working. I'm asking if anyone of you had a close problem who may help me. Thanks
